Un groupe de recherche de Kiel a d\u00e9couvert un m\u00e9canisme qui pourrait avoir un impact sur cette approche.<\/strong><\/p>\n<p> <!--more--> <\/p>\n<p>Normalement, le messager interleukine-22 (IL-22) a pour fonction de soutenir la r\u00e9ponse immunitaire intestinale aux infections pathog\u00e8nes et de favoriser la cicatrisation de l&#8217;\u00e9pith\u00e9lium. Il le fait par la prolif\u00e9ration cellulaire et l&#8217;induction de prot\u00e9ines antimicrobiennes. Il aide ainsi la muqueuse intestinale, endommag\u00e9e par l&#8217;inflammation, \u00e0 se r\u00e9g\u00e9n\u00e9rer. Des prot\u00e9ines de fusion recombinantes de l&#8217;interleukine 22 sont actuellement d\u00e9velopp\u00e9es pour aider les patients atteints de maladies inflammatoires chroniques de l&#8217;intestin.<\/p>\n<p>Mais l&#8217;IL-22 peut aussi agir diff\u00e9remment, comme l&#8217;ont d\u00e9couvert des chercheurs du cluster d&#8217;excellence de recherche sur l&#8217;inflammation de Kiel. A savoir, lorsque les patients sont d\u00e9pourvus du g\u00e8ne ATG16L1. Cela concerne tout de m\u00eame un cinqui\u00e8me des patients atteints de Crohn.<\/p>\n<p>ATG16L1 joue un r\u00f4le important dans l&#8217;autophagie. L&#8217;autophagie est responsable de la d\u00e9gradation intracellulaire des prot\u00e9ines et organites cellulaires d\u00e9fectueux ainsi que des agents pathog\u00e8nes, d&#8217;o\u00f9 son importance pour la r\u00e9ponse immunitaire. Si ATG16L1 est absent, l&#8217;autophagie est perturb\u00e9e. La d\u00e9gradation est plus lente et les inflammations plus fr\u00e9quentes. Le comportement de l&#8217;IL-22 change \u00e9galement : lors d&#8217;une exp\u00e9rience sur des souris, on a constat\u00e9 que l&#8217;IL-22 agissait de mani\u00e8re contraire \u00e0 son comportement normal chez les souris d\u00e9pourvues d&#8217;ATG16L1. Au lieu de favoriser la r\u00e9g\u00e9n\u00e9ration cellulaire, il a un effet pro-inflammatoire.
